Cytotoxic Activity of Zerumbone-Loaded Chitosan Oleic Acid Nanoparticles

Zingiber zerumbet contains zerumbone has high anticancer activity on various cancer cell lines. However, zerumbone has
low bioavailability due to its low solubility in water. In order to increase bioavailability, zerumbone formulated by
nanoencapsulation. The purpose of this study was to assess the cytotoxic effect of zerumbone nanoencapsulated with oleic
acid-modified chitosan. Characterization of encapsulated zerumbone was conducted using Particle Size Analyzer.
Cytotoxic activity on T47D cells was evaluated by MTT assay method. The results showed that the nanoparticles of
zerumbone were in the nano range (~500 nm) and the zeta potential were ~30 mV. Zerumbone with chitosan and
nanoparticle zerumbone with chitosan-oleic acid could inhibitthe growth of T47D breast cancer cells with IC50of 58.91 and
45.65 μg/mL, respectively. Cytotoxic mechanism of those are apoptotic induction and cellular necrosis.
